Applies technical expertise and experience to plan, execute, and analyze experiments that advance Hart Lab research goals in the department of Bioinformatics and Computational Science. The research plan involves using high-throughput multiplex genetic screens to explore the dynamics of genetic interaction networks and discover synthetic lethalities for therapeutic exploitation.

JOB SPECIFIC COMPETENCIES

In vitro genetic screens. Designs and acquires CRISPR reagents, typically oligonucleotide pools, and clones pools into relevant lentiviral vectors, and sequence verifies products. Lentiviral packaging and titering for pooled library screens at specified MOI. Selection, maintenance, and accurate quantitation of transduced cells using good sterile technique.

High-throughput sequencing. Prepares custom libraries for multiplexed sequencing on an Illumina and/or Nanopore platform. Collection of genomic DNA, amplification of CRISPR gRNA cassettes, library preparation. Accurate measurement of sequencing library purity and concentration for sample multiplexing. Maintain knowledge of current sequencing technologies and improve lab protocols/methods as appropriate.

Data Management and Analysis. In coordination with laboratory informatics staff, manages deposition of quantitative screen data (sequence or cytometry) in institutional data storage. Provides preliminary quality control review and analysis of screen data. Works with informatics staff to identify, develop, and apply existing or novel algorithms to extract biological meaning from screen data.

Validation experiments. Design and perform, or supervise the performance of, validation experiments using a broad array of genetics, molecular biology, and biochemistry techniques to validate observations/predictions gleaned from screen data.

Communication, lab organization, and record keeping. Maintains accurate and up to date lab notebooks. Establishes and maintains laboratory standard operating practices for efficient screening and sequencing, within designated safety regulations. May plan and supervise the training of technicians and other laboratory personnel with respect to proper laboratory techniques, the use of laboratory equipment and safety procedures. Periodically reviews and updates protocols. Interacts with all levels of internal and external resources to achieve lab goals and resolve problems. Share protocols and best practices with internal and external collaborators. Communicate research goals, progress, obstacles points in quarterly lab staff meetings and in ad hoc scientific meetings as appropriate.

EDUCATION:
Required: Masters degree in one of the natural sciences or related field.

EXPERIENCE:
Required: Four years of experience in experimental or scientific work. With PhD, no experience required. With Medical degree, two years of experience in experimental or scientific work.
