Position Summary:
The Cell Engineering group is recruiting a creative and highly-motivated scientist to join the Design Biology team as a subject matter expert (SME) in high-throughput (HTP) strain development. The successful candidate will develop innovative methods to build strains containing pooled, combinatorial DNA parts for yeast and bacteria while using state-of-the-art genetic engineering technologies. The goal of these complex strain libraries is to enable the rapid identification of advantaged genotypes for the bioproduction of small molecules using top-down high-throughput phenotypic screens. This team member will collaborate closely with SMEs in bioinformatics and automation while working with multiple bio-molecule project leaders to deliver strain libraries with desired specifications. This is a critical role that will enable a step change in strain design capability within ADM Cell Engineering. Scientists with experience working with industrially relevant non-model microorganisms are encouraged to apply.
Responsibilities:
- Develop and use state-of-the-art synthetic biology methods for building complex DNA variant libraries (e.g. enzyme variants, multigenic expression cassettes) in microbial genomes
- Collaborate with the high-throughput automation team to ensure strain libraries fit specifications and that DNA parts are catalogued in LIMS
- Work with molecule project leaders to design and deliver genetically diverse libraries that meet strain KPI improvement goals
- Develop technologies to standardize strain engineering methods that increase efficiency measured by true positive rate (TPR) of strains identified from screens
- Determine the technical feasibility and execute strain library builds with increasing complexity (e.g. multi-enzyme pathways)
- Expand and optimize existing workflows for improved economics, success rate and turnaround time
- Assist with the bench-to-robot transfer of the technology developed
- Simultaneously work on multiple strain projects with different specifications
- Track genetic parts and strain designs using a customized LIMS system
- Contribute to a team-oriented, positive, safe and inclusive work environment
These skills and experiences will be required for the candidate:
- Ph.D. in molecular biology, biotechnology, biochemistry or related field with 3+ years of industry experience
- Track record developing high-throughput molecular biology workflows that were transferred to automated platforms
- Experience developing novel methods and troubleshooting molecular biology protocols to build large strain libraries
- Deep understanding of state-of-the-art genome editing technologies
- Experience working with large strain datasets derived from HTP screens
- Experience genetically engineering non-model industrial microorganisms including extremophilic yeast and/or bacteria
- Demonstrated success working within inter-disciplinary teams
- Excellent written and oral communication skills with both technical and non-technical personnel
- Ability to prioritize responsibilities and work effectively in a matrix environment
