DEPARTMENT DESCRIPTION:
The job position is part of the Upstream team in the drug substance function within global Manufacturing Science & Technology (MSAT) and is based at the biologics hub in Framingham, MA. The MSAT mission is to enable the reliable supply of SC (Specialty Care) medicines by providing expert technical and scientific support to their licensure, ongoing commercial manufacturing, and life-cycle management activities.
The drug substance function within global MSAT is the owner of Life Cycle Management (LCM) of DS processes within the Sanofi biologics manufacturing network. We deliver innovative, robust, and cost-effective next generation processes and provide commercial manufacturing support. The function is responsible for tech transfer activities including process-fit gap assessment, process validation, dossier preparation, & PAI support to enable launch of new and LCM products. Our expansive portfolio covers 17 products, 5 modalities, 3 expression systems within 12 internal and external global sites in the US and EU. We will be supporting launch of 20 new products in the next 5 years including monoclonal and multi-specific antibodies, fusion proteins, antibody-drug conjugates, synthorins, and nanobodies. In recent years we have successfully established second-generation manufacturing processes with a continuous process platform. We are pursuing future innovations such as digital labs, factory of the future, and advanced-analytics-based process understanding and control.
Job Responsibilities
Support commercial biotech manufacturing of mammalian cell culture and protein purification through experimental work and data analysis to support scale-up, mixing & mass transfer analysis, and process analytical technology.
Work as part of a team, engaging other scientists with complementary skill sets in the field of Biopharmaceutics
Generate documents (technical memos, protocols, reports, etc.) related to lab experiments.
Co-ops will own a relevant capstone project to complete during the semester.
Basic Qualifications
Must be currently enrolled as a student in an accredited undergraduate or graduate academic program
Working toward a bachelor's or master’s degree in Chemical Engineering, Biochemistry, or equivalent
3.0/4.0 GPA is a minimum requirement
Must have permanent US work authorization
Preferred Qualifications
Basic understanding of mammalian cell culture
Understanding of mass transport and building of system boundaries for optimization
Experience in scripting in Matlab or Python
Knowledge in Chemometrics and multivariate data analysis
Co-Op Project Description
Develop dynamic kLa models for small-scale bioreactors
The mass transfer coefficient, kLa determines the rate of gas transport between gas and liquid phases, and the method is used for oxygen uptake rate measurement and the prediction of oxygen demands. However, the current kLa measurement methodology is not accurate due to the high contribution of surface kLa in small-scale bioreactors, but the surface kLa is assumed as it is negligible. Here, we will develop
Direct measurement of headspace DO (dissolved oxygen) as well as liquid phase DO in a small-scale bioreactor.
Build a dynamic kLa model using headspace DO and liquid phase DO and estimate surface kLa and liquid phase kLa in a small-scale bioreactor.
Apply and compare classic (apparent) kLa and dynamic kLa (liquid and surface kLa) using cell culture simulation.
Develop Raman spectroscopy models to predict metabolite concentrations and cell density using the Ambr250 (high throughput and miniaturized bioreactor), Raman PAT and Simca (MVDA analysis software). The Ambr Raman PAT models can be developed efficiently using the high throughput operations of Ambr250 and to minimize the model development time.
The candidate will learn the Ambr250 operation for the Raman PAT and optimize Raman model parameters for metabolites and viable cell density in the mammalian cell culture process.
